there are 22 test chambers/levels if you don't count the other parts of the plot

it is not actual chembers like portal 1,you have to go through numbers of puzzles to progress,so they are not the Chembers from portal 1,later in game you have to solve like 17/18 rooms like portal 1,thats it,so there is no actual number of chembers in portal 2

Apparently there are 22 chambers/levels with many other plot sections and areas. the developers promised atleast 10 hours of gameplay. Plus, there's the Co-op levels.
